I began with the idea of the Gods being the largest characters in sensei Sakai's work.
When i looked at the drawings i realized there was no scale with which to measure the Gods on the first few pages of Grasscutter.
A few pages after the Gods is the 8-forked serpent of Koshi on page 15 of Grasscutter.
These were good answers, but Steve's is ichiban!!!!!!!!!!!!

And Lord Hebi would be the tallest of all the characters whom Stan Sakai actually created….

I am going to say that Usagi Gundam from Usagi Yojimbo: Senso is the largest character and
I will guess Usagi Gundam is as tall as 50 Usagi Yojimbos

Usagi Gundam is as tall as 20 Usagi's.
Assuming Usagi is a 5 foot tall bunny, then Usagi Gundam has to be at least 100 feet tall.
Not sure how the ears fit into this equation, but that's my guess.

My guess is Lord Hebi. I'm thinking he's 4 or 5 Usagi's long/tall.
I am also happy with using Usagi as a unit of measurement, just sounds right.

When you say largest character in Sensei Sakai's work,
do you mean anything he's draw (even outside the Usagi universe)?
I'd wager that King Kong is one of the largest. In the "Kong of Skull Island" universe, Kong measures about 100'-120' tall.
I figure that Usagi, with ears (not pointed straight up), is about 6' tall.
That would make Kong approximately 17-20 Usagi's tall.
Perhaps even larger is Usagi Gundam.
I made some measurements of the drawings and figure that UG is about 30-34 Takenoko-Sensei's high.
Usagi is taller, so I'd imagine that UG is about 27-30 Usagi's tall.
I'm not trying to win, but thought it would be fun to participate in this challenge.

I'll launch with an obvious one first to see how I get on... Usagi Gundam,
which when measuring the page in question, appears to be about 20 Usagis high, or more, as you only get up to its top knot on the page!

You never said that the character had to be in the Usagi universe so I will throw out another guess of Kong is the largest character and I will guess

How about Kamakura Akugenta as a stone majin giant? 7 and a half Usagi's?
or Godzilla from Sumi-e part 3. Also around 7 and a half Usagi's.
Can't quite tell which is taller based on how Stan draws them.
They are much smaller than the Usagi Gundam (vehicle).

My guess is the Kamakura Akugenta Stone Majin Giant in Sumi-e Part 3, Usagi Yojimbo V3, issue 68.
This character is as tall as 10 Usagi Yojimbos (my estimate).

My final guess will be the octopus demon from My Lord's Daughter.
When Usagi is riding the shark to the surface, you can see that the tentacles alone have to be 10 or 11 Usagi's tall.
So I'll go with 15 Usagi's since I can't see the head.
