Back in 1991, Stan Sakai did an interview for Amazing Heroes, a magazine put out by Fantagraphics, Stan's publisher at the time. Included in that magazine was an eight page comic titled "Portrait of the Artist as a Young Cartoon Character" which described the creation of an issue of Usagi Yojimbo from beginning to end.
A few years later, that story was revised and expanded with 4 new pages detailing how a cover is create, and published in the first issue of The Art of Usagi Yojimbo published by Radio Comix in 1997. The title was changed to "How I Create an Issue of Usagi Yojimbo"
Now, just a few years ago, Dark Horse Comics released their large 20th Anniversary Art of Usagi Yojimbo volume which included a reprint of that story. If you go to the Amazon.com page for that book, you can read most of that story right on the web-site.

Another option is this video taken at one of Stan's library presentations where he gives a step by step description of his cteative process.

I'm pretty sure it was in Amazing Heroes #187, with Usagi on the cover. It was expanded and reprinted in UY Book 5: Lone Goat and Kid hardcover. I think it was also in The Art of UY comic from Radio Comix. It was again reprinted in The Art of UY book from Dark Horse.Mark Bordiyan wrote:And do you know what number of the magazine that is!? I also did a step-by-step look at how I create a story in the UY Sketchbook #3. UY Sketchbook #7 has a short look at the process behind the 3-page Netsuke story originally done for Wizard Magazine.

Konichiwa!! I have that issue and it's a treasure trove of information! It's also repeated in "The Art Of Usagi Yojimbo #1"comic. I of 2 issues.
